KLF6在肝细胞癌中的表达及在肝癌细胞系中对增殖和凋亡的影响

摘要 目的检测抑癌基因KLF6在人肝细胞癌中的表达情况,探讨KLF6在肝癌细胞系中对细胞增殖及凋亡的影响。方法分别用荧光定量PCR、RT-PCR和Western blot、免疫组织化学方法检测肝细胞癌、癌旁组织及正常肝组织中KL F6基因mRNA及蛋白水平的表达情况。构建KLF-6基因的真核表达质粒,用MTT法检测细胞的增殖情况,镜下凋亡荧光染色法检测细胞凋亡情况。结果 KLF6基因mRNA和蛋白在肝癌组织中的表达水平明显低于配对的癌旁组织和正常肝组织,差异有统计学意义(P=0.001)。与pcDNA3.1(-)空质粒转染组及未转染组细胞相比,KLF6转染组的细胞生长受到抑制,细胞凋亡明显增加。结论 KLF6基因通过抑制肝癌细胞增长、增加肝癌细胞凋亡,在肝细胞癌的发病机制中发挥重要作用。 Objective To detect the expression of KLF-6,a tumor suppressor gene,in human hepatocellular carcinoma and to explore its role in proliferation and apoptosis in HCC cell lines.Methods The expression of KFL6 mRNA and protein were tested by Real Time PCR,immunohistochemistry and Western blot respectively in hepatocellular carcirnoma tissues,and paracancerous tissues and normal liver tissues.Corresponding plasmid of KLF6 was conducted,and proliferation of target cells was measured with MTT.Results Compared with paracancerous tissues and normal liver,KLF6 mRNA and protein were over expressed in hepatocellular carcirnoma(P=0.001).When compared with pcDNA3.1(-)empty vector transfected cells and untransfected cells,the proliferation of KLF6 transfected cells was inhibited and apoptosis of transfected cells increased significantly.Conclusion KLF6 gene plays an important role in the pathogenesis of HCC by inhibiting the growth of HCC cells and increasing the apoptosis.
