摘要
目的:为寻找防治药物依赖新方法,本文以大鼠条件性位置偏爱(CPP)为模型,探讨伏隔核电刺激对阿片类物质奖赏效应的影响。方法:20只成年大鼠分为两组:深部脑刺激(deep brain stimulation,DBS)组和假DBS组。用立体定位手术将DBS装置的电极部分植入一侧伏隔核(NAc)核心部,与脉冲发生器(IPG)连接后,一起固定于大鼠颅顶。一周后以递增剂量腹腔注射(ip)吗啡,产生位置偏爱后给予DBS组130 Hz高频电刺激(HFS),每日1次。结果:电刺激14 d后DBS组未产生条件性位置偏爱再现,假DBS组产生条件性位置偏爱。结论:伏隔核高频电刺激能够抑制大鼠吗啡CPP的再现。
Objective: To explore a novel method for the treatment of drug dependence,we investigated the effect of deep brain stimulation(DBS)of nucleus accumbens(NAc)on the rewarding effects of opiates,using morphine conditioned place preference(CPP)in rats.Methods:Twenty adult rats were divided into two groups,DBS group and sham DBS group.The DBS electrode was stereotaxically implanted into the core of unilateral NAc,connected to an implantable pulse generator(IPG),and they were fixed to the rat skull.One week later,the rats in each group were intraperitoneally injected with morphine at an increasing dose until the CPP appeared.Then,the rats in the DBS group were administered a 130-Hz high-frequency stimulation(HFS)once daily.Results:After 14 d administration of electrical stimulation,rats in the sham DBS group showed significant CPP,while rats in the DBS did not.Conclusion:The HFS of the rat NAc may attenuate the reinstatement of morphine CPP.
