摘要
目的研究N-钙粘素(N-cadherin)、碱性成纤维生长因子(bFGF)、增殖细胞核抗原(PCNA)在高温致神经管畸形神经上皮组织中的表达及其意义。方法在高温致金黄地鼠神经管畸形的动物模型上,用免疫组织化学法检测不同时间实验组和对照组鼠胚神经管上皮细胞N-cadherin、bFGF、PCNA的表达,并进行定量和定位的观察。结果 (1)高温处理后,16 h和24 h实验组N-cadherin在神经管上皮细胞的表达低于对照组(P<0.05),N-cadherin在增殖旺盛的神经上皮表达稳定;(2)高温处理后,24 h和48 h实验组bFGF在神经管上皮细胞的表达低于对照组(P<0.05),2组bFGF的表达随胚龄增长有增加的趋势。(3)高温处理后,162、4 h和48 h实验组PC-NA在神经上皮的表达弱于对照组(P<0.05),2组阳性反应范围均有所缩小。结论高温通过影响N-cadherin、bFGF、PCNA的表达,诱导神经管畸形的发生。
Objective To study the expression and signification of N-cadherin,bFGF,PCNA in the neuroepithelium of neural tube defects induced by hyperthermia.Method The expression of N-cadherin,bFGF,PCNA in the neuroepithelium were examined quantitatively with immunohistochemistry in the golden hamster NTDs model.Results Firstly,After 16 and 24 hours exposure,the level of N-cadherin in the neuroepithelium was lower than control group(P〈0.05),especially at the 24 th hour(P〈0.01),N-cadherin still kept positive on the neuroepithelium with proliferation;Secondly,the amount of bFGF was significantly lower than control group after 24 h and 48 h(P〈0.05).The expression of bFGF gradually increased accompanied by gestational age in both experimental and control groups;Thirdly,the content of PCNA of the epithelial cells in the experimental groups decreased after16 hours(P〈0.05),by the 48 th hour,the field of positive reaction in neural tube narrowed in both two groups.Conclusion It is suggested that the expression of N-cadherin,bFGF,PCNA maybe play a role in the pathogenesis of neural tube defects intervened by hyperthermia.
