基于抗菌导向的乳铁蛋白异源表达研究进展

Advances on Heterogenous Expression of Lactoferrin Towards Antimicrobial Role

摘要 乳铁蛋白是哺乳动物天然免疫系统和获得性免疫系统中的重要防御成分,具广泛生物学功能,包括调节体内铁平衡、广谱抗菌、抗炎症、抑制肿瘤生长、增强机体免疫力等,在医药、食品、饲料领域有重要应用价值。目前乳铁蛋白规模化生产技术瓶颈是提取成本高,利用基因重组技术构建高效表达系统是突破这一瓶颈的重要途径。基于抗菌导向的乳铁蛋白及其衍生分子在大肠杆菌、酵母、昆虫、哺乳动物和植物中表达的研究进展进行了综述。 Lactoferrin is a key component of the mammalian immune defense system.It shows a multifunction involving in iron regulation,broad spectrum of antimicrobial activity,anti-inflammatory activity,anti-tumor activity and immunologic enhancement.But its potential nutritional and therapeutic applications have been limited due to the high cost for extraction and purification of functional lactoferrin from milk.Recombinant expression would be a feasible approach to produce active lactoferrin in large scale.The advances of lactoferrin recombinant expression in E.coli,yeast,insect,plant and mammalian cells were reviewed.
