脂蛋白电泳结合酶显色测定血清各脂蛋白中甘油三酯及临床应用 被引量:5

Determination of triglyceride in lipoproteins separated by agarose gel electrophoresis and its clinical application

摘要 采用琼脂糖脂蛋白电泳分离脂蛋白结合甘油三酯(Tg)酶试剂呈色,扫描出各种脂蛋白区带中Tg所占比例,同血清总Tg浓度相乘计算各脂蛋白组份中Tg的含量。结果示本法与参考法比较相关系数:HDL-Tg、VLDL-Tg、LDL-Tg分别为0.873、0.955、0.912。方法的批内CV:HDL-Tg、VLDL-Tg、LDL-Tg分别为8.0%~10.2%、3.8%~6.5%、4.3%~7.0%;批间CV分别为9.5%~16.2%、6.8%~8.5%、4.8%~9.7%。平均回收率:HDL-Tg、VLDL-Tg和LDL-Tg分别为:88.8%、93.4%和91.7%。冠心病组同相应的对照组比较LDL-Tg(0.50±0.32vs0.28±0.09mmol/L)及LDL-Tg/LDL-Ch比值(0.211±0.316vs0.095±0.033)均显著升高。该法简便易行,是精密准确定量各脂蛋白中Tg的理想方法。LDL-Tg及LDL-Tg/LDL-Ch比值为冠心病有价值的诊断指标。 A simple method was developed for measurement of triglyceride(Tg)in lipoproteins. Lipoproteins separated by agarose gel electrophoresis.The Tg of bands was stained by specific enzymatic method(GPOTrinder)after electrophoresis.The relative amounts of Tg in lipoproteins were determined by densitometric scanning at 520 nm. Absolute triglyceride concentrations of the respective lipoprotein fractions were calculated from total Tg. Comparison of the electrophoresis method with a combined ultracentrifugation/precipitation method in 24 serum resulted in following correlation coefficients:HDLTg:=0873;VLDLTg:=0955;LDLTG:=0912The intraassay and interassay coefficients of variation were:8010238654370%;9516268854897%The average recovery was 888%934%917% respectively.This method is easy to perform,and is a precise and accurate technique for the determination of lipoprotein Tg.
