目的:细胞核仁形成区( N O Rs)主要由核糖体 R N A 基因组成,位于此区调控 r R N A 转录的酸性非组蛋白可被硝酸银染色( Ag N O Rs), Ag N O Rs 变化反映了细胞代谢调控异常及细胞功能受到影响。本研究观察海洛因依赖患者外周血 T 淋巴细胞 Ag N O Rs 及与解毒有关的胎盘型谷胱甘肽 S转移酶( G S Tπ)表达,分析海洛因依赖对 T 细胞代谢乃至功能的影响。方法:利用北京开隆公司提供的 K L 型免疫图像分析系统检测 T 细胞 Ag N O Rs;通过定量 P C R 技术分析患者外周血 G S Tπ表达。 结果: 72 例海洛因依赖患者外周血 T 淋巴细胞 Ag N O Rs 检测平均值 (6.78 ±0.50) % ,明显低于正常人,但高于肿瘤患者;其中 33 例进行了 G S Tπ表达分析,结果表明,与正常人比较海洛因依赖患者外周血 G S Tπ表达明显增高;且 Ag N O Rs 与 G S Tπ表达呈明显负相关。结论:海洛因依赖患者外周血 T 淋巴细胞 Ag N O Rs 明显降低,表明其免疫细胞功能受到明显抑制。 Ag N O Rs 降低的幅度明显小于肿瘤患者,说明海洛因依赖对 T
Objective: To analyze the expression of nucleolar organizer region (NORs) associated proteins(Ag NORs) in heroin dependence individuals using the KL Imaging System and to compare the results with those of the control group and cancer patients. Methods: 72 cases of heroin dependence were analyzed with KL Imaging System, while the expression of glutathione S transferase pi(GST π) in 33 cases of heroin dependence was measured by quantitative PCR. Results: The Ag NORs in heroin dependence individuals were (6.78±0.50) % in average which is lower than that in control group with statistic significance. Both the expression rate and level of GST π were significantly higher in heroin dependent cases than in normal control, but no relationship between the GST π mRNA and sex, dependence time or the type of drug taking was found. In addition, there was negative correlation between Ag NORs and GST π. Conclusion: Heroin dependence might induce immune damage with decreased Ag NORs in T cells and increased expression of GST π in blood cells.
