Establishment and evaluation of an asthmatic mouse model

摘要 目的探讨哮喘小鼠模型建立的方法。方法将20只SPF级BALB/C雌性小鼠随机分成对照组和模型组两组。模型的构建采用腹腔注射OVA+氢氧化铝凝胶致敏3次后予1%OVA雾化激发7 d,以生理盐水腹腔注射及雾化吸入为对照。最后1次雾化激发1 h后眼球取血,取右肺作HE染色;左肺收集BALF,进行细胞计数;ELISA检测外周血IL-12水平。结果模型组小鼠激发可见典型的哮喘症状。模型组小鼠BALF中炎性细胞总数和EOS较对照组明显增多,外周血IL-12水平显著降低,且IL-12水平与BALF中EOS数呈高度负相关。结论致敏3次后雾化激发是一种较好的建立哮喘小鼠模型的方法。 Objective To investigate the method in establishing asthmatic mouse model. Methods 20 SPF female BALB/C mice were randomly divided into control group and model group. We The mice in asthmatic guoup were injected intrapefitonealy with OVA 3 times and aluminum hydroxide gel to be sensitized and excited with 1% OVA spray The control group were treated with saline as substi- tution of OVA. Mice were sacrificed by cervical dislocation and the peripheral blood were obtained at 1 h after the last aerosol exposure, the right lungs were deligated. And the left lungs were lavaged, the number of inflammatory cells in the mouse bronchoaleolar lavage fluids (BALF) were counted with microscope. Using enzyme-linked immunosorbent assay(ELISA) to detect levels of IL-12 in serum. Results We can see typical symptoms in model group. In model group, the total number of inflammatory cells and EOS significantly increased in BALF, levels of IL-12 significantly reduced in serum. Levels of IL-12 in serum had altitude negative relationship with the EOS counts in BALF (r = - 0. 776, P 〈 0. 01 ). Conclusion 3 times sensitization and excited spray with OVA, is a better method in establishing asthmatic mouse model.
