摘要
分别利用戊二酸酐法和碳化二亚胺法合成半抗原和免疫抗原免疫动物,制备特异性强的单克隆抗体。在筛选硝基咪唑类单克隆抗体的基础上,建立ELISA检测方法,并研制出对鸡肉中硝基咪唑类药物残留检测试剂盒。试剂盒最低检测限为0.04μg/kg,样本添加回收率为69.4%～107.5%,批内变异系数为7.8%～14.6%,批间变异系数为10.9%～16.5%。该方法的建立为硝基咪唑类药物残留的检测提供了便捷、准确的分析检测手段。
The glutaric anhydride and carbodiimide coupling methods were used to synthesize hapten and immunogen respectively. Specific monoclonal antibody was produced by animal immunity. On the basis of nitroimidazoles monoclonal antibody,rapid test ELISA kit for nitroimidazoles residues in chicken was developed. The detection limit of ELISA kit was 0.04 μg/kg. Sample recoveries of chicken samples were 69.4% to 107.5%. Intra C.V. of chicken samples were 7.8% to 14.6%. Inter C.V. of chicken samples were 10.9% to 16.5%. This method provided a convenient and accurate way for detection of nitroimidazoles residues.
