Preparation of Metformin enteric sustained release microcapsules by spray drying

摘要 目的采用喷雾干燥法制备二甲双胍肠溶缓释微囊并对其体外释放度进行考察。方法以包封率为评价指标,对喷雾干燥工艺参数进行正交优化设计,并考察Eudragit L100用量、微囊芯材比对微囊性能的影响。结果最优工艺条件为进风温度165℃;进料速度10 mL·min-1;喷雾压力0.3 MPa;药物与Eudragit L100材料的质量比1∶3;制得的微囊外形良好,包封率可达96.38%;在人工肠液中缓慢释放,药物1 h释放量不超过30%,24 h不低于99%。结论所得制备工艺可行,制得的微囊球形圆整,包封率较高,具很好的缓释特征。 OBJECTIVE To prepare metformin enteric microcapsules by spray drying and study its in vitro drug release characteristics.METHODS Orthogonal designing was applied to investigate and optimize the technique operation of spray drying.The entrapment rate(ER) used as an index,the influence of the amount of Eudragit L100 and the ratio of core and Eudragit L100 on microcapsules characteristics were studied.RESULTS The best condition for spray drying technique was as follows:temperature was at 165 ℃;feeding rate was 10 mL·min-1;spray pressure was 0.3 MPa;the best ratio of core and polymer material was 1:3.ER of microcapsules could reach up to 96.38%.The drug release was no more than 30% in 1 h,no less than 99% in 24h.CONCLUSION The metformin microcapsules prepared by the established method were round with satisfied ER and slow release.
