摘要
用培养的新生鼠心肌成纤维细胞, 加血管紧张素Ⅱ(AngⅡ) 孵育, 考察引起心肌肥厚的生长因子AngⅡ对胶原基因转录水平的影响。RT-PCR结果显示AngⅡ可刺激胶原Ⅰ、Ⅲ型m RNA表达水平显著增加, 此作用可被AT1受体拮抗剂芦沙坦 (Lorsatan) 所抑制。
Although the collagen subtypes Ⅰ(Co Ⅰ)and Ⅲ(Co Ⅲ) are essential components of the cardiac extracellular matrix (ECM) maintaining the functional integrity of the heart, little is known of the temporal relationship between events in collagen gene transcription and the occurrence of cardiac fibrosis ,or of the regulation of these events by angiotensin (AT 1) receptors. The aim of the present study was to investigate whether angiotensin Ⅱ (Ang Ⅱ)stimulation known to induced cardiac hypertrophy might be involved in the regulation of the collagen gene, neonatal cardiac fibroblast (NCF) cultures were prepared from Sprague-Dawky rats 1-3 d after birth. Ang Ⅱ was used as a hyperplastic agent. Collagen Ⅰ and Ⅲ type mRNA expression was detected by RT-PCR. Stimulation of NCF with 1 μmol/L Ang Ⅱ for 24 h resulted in a significant increase of the collagen I and collagen Ⅲ mRNA expression,which could be inhibited by the AT1 receptor antagonist losartan.
