摘要
目的利用组织浴槽的方法体外模拟在体桡动脉,摸索建立体外桥血管药物敏感模型,研究单一因素改变对血管舒缩状态的影响。方法取成年男性桡动脉,4℃Krebs-Ringer保存液中切割成3~4 mm宽、内膜完好的血管环,两端分别牵引,在10 ml麦式组织浴槽中水浴,并连接至换能器、生理记录仪和适度的前负荷上,待血管环状态稳定后记录初长度及静息张力。分别用终浓度为0.06 mol/L的K+和20μmol/L的乙酰胆碱评估内膜组织的完整性和证实血管环舒张、收缩活性。取内膜完整且舒缩活性良好的血管环分别用不同终浓度的苯肾上腺素作用作为刺激因素,观测血管环收缩程度的变化。绘制在不同半对数克分子浓度苯肾上腺素作用下,血管环收缩静息张力变化曲线。结果所有血管环在水浴槽中均能很快达到稳定状态。对KCl和乙酰胆碱的反应敏感,提示血管环内膜完整及收缩、舒张功能良好。苯肾上腺素半对数克分子浓度与血管环静息张力呈正相关。结论利用组织浴槽建立药物敏感模型在方法上是可行的。血管环可以很好的模拟桥血管在体内的状态,可将实验所得苯肾上腺素浓度-静息张力曲线中的半对数克分子浓度作为后续实验中刺激血管收缩的标准浓度。
Objective To establish a drug sensitive model for radial artery in vitro by utilizing the organ bath to mimic radial artery in vivo,and to study the unifactor which affects the systolic and diastolic state of the radial artery.Methods The radial arteries were harvested from adult male and cut into 3 to 4 mm vessel rings with intima intact in the Krebs-Rubger liquid at 4 ℃.The vessel ring was pulled with proper preload at both ends and connected to the pressure transducer and physilogical recorder in the organ bath.The initial length and resting tension were recorded after stabilization of the radial artery.The 0.06 mol/L KCL and 20 μmol/L acetylcholine were used to evaluate the integrity of the intima and the systolic and diastolic activity of the blood vessel.The vessel ring which had fine integrity of intima and systolic and distolic function were tested with different concentrations of phenylephrine to obtain the changes of systolic and diastolic extent.The systolic resting tension curves were drawn at different semi-logarithmic drug molar concentrations.Results All the vessel rings reached steady state at a short time in the organ bath.The sensitivity to the KCl and acetylcholine indicated that the intima integrity and systolic and diastolic function of the vessel ring were fine.There was significantly positive correlation between semi-logarithmic drug molar concentration of phenylephrine and resting tension of blood vessel ring.Conclusion It is feasible to utilize the organ bath to establish the drug sensitive model for radial artery in vitro.The radial artery ring mimics the state of the artery graft in vivo with great satisfaction.The semi-logarithmic drug molar concentration obtained from the phenylephrine concentration and resting tension curve could be employed as standard concentration for the following testing of blood vessel systolic function.
