神经干细胞移植对大鼠脑损伤β-catenin的表达的影响

摘要 目的通过观察神经干细胞(NSCS)移植治疗大鼠脑损伤前后大脑皮质神经元β-连环蛋白(β-catenin)的表达变化,探讨NSCS移植治疗对脑外伤功能恢复的作用。方法自由落体撞击大鼠左脑大脑皮质运动感觉区以制作脑外伤模型,移植培养的NSCS,采用免疫组织化学、免疫印迹方法检测脑外伤后2w和4w及脑外伤神经干细胞移植后2w和4w时细胞骨架蛋白β-catenin的表达变化。结果创伤组β-catenin的表达水平比假手术组减少,创伤后移植组β-catenin的表达水平比创伤组增加(<0.05)。结论 NSCS移植对脑外伤的修复作用可能与β-catenin的表达有关。 Objective To investigate the effect of transplantation of neural stem cells(NSCS) on β-catenin protein expression in cortex of SD rat with brain weight-dropping injury. Methods NSCS were cultured and transplanted into SD rat cortical sensorimotor area injured by weight-dropping within 24 hours. Immunohistochemistry and western blotting were used to detect the expression of β-catenin. Results The expression level of β-catenin protein in the cortical sensorimotor area of rats was decreased in the injuried group than in sham group,but the expression level of β-catenin was increased in NSCS transplantation group than in the injuried group( 0.05) . Conclusion The repair effect of NSCS on the brain injury might be related to the expression of β-catenin.
