三羟异黄酮对去卵巢大鼠学习记忆及海马神经元细胞凋亡的影响 被引量:1

Effects of Phytoestrogen on Learning and Memory in Ovariectomized Rats and the Underlying Mechanism

摘要 目的:观察三羟异黄酮(GST)对去卵巢大鼠学习记忆能力及海马神经元细胞凋亡影响,探讨植物雌激素对中枢神经系统的保护作用及可能作用机制。方法:选取成熟雌性清洁级大鼠50只,随机分为假手术组,去卵巢对照组,GST低、中、高剂量组,每组10只。术后1周,GST低、中、高剂量组大鼠每天灌胃以二甲基亚砜为溶媒的GST溶液(剂量分别为25、50、100mg/kg体质量);去卵巢对照组和假手术组大鼠每天灌胃与受试药物等容积的溶媒二甲基亚砜。实验持续4周。水迷宫实验观察各组大鼠学习记忆能力,流式细胞液检测大鼠海马神经元凋亡;结果:与假手术比较,去卵巢对照组大鼠水迷宫学习记忆能力潜伏期明显延长(P<0.05);与去卵巢对照组比较,GST低、中、高剂量组大鼠水迷宫学习记忆能力潜伏期明显缩短(P<0.05),但各实验组间比较,差异无统计学意义(P>0.05);去卵巢对照组海马内Annexin V含量较假手术对照组明显升高(P<0.01),GST低、中、高剂量组与去卵巢对照组比较,Annexin V表达明显降低(P<0.05);GST低、中、高剂量组间比较,及与假手术组比较,差异无统计学意义(P>0.05)。结论:三羟异黄酮能明显改善去卵巢大鼠学习记忆能力,其作用机制可能与抑制去卵巢大鼠海马神经元细胞凋亡有关。 Objective: To investigate the effects of genstein,a kind of phytoestrogen(PE) derived from a soy diet,on capabilities of learning and memory in ovariectomized rats,and to explore the underlying mechanism.Methods: Fifty female Sprague Dawley rats were randomly divided into 5 groups: sham-operation group,ovariectomized group;and low(25 mg/kg),medium(50 mg/kg) and high(100 mg/kg) doses of genstein groups,10 in each.Three genstein groups were given 25,50,and 100 mg/kg genstein in 1 week after operation,respectively,and the sham-operation and ovariectomized groups were given equivalent volume of the vehicle in which genstein was dissolved.After 4-week treatment,Morris water maze test were used to observe the ability of learning and memory.The number of apoptosis neurons in the hippocampus was detected by flow cytometry.Results: The ovariectomized group had significantly prolonged average latency as compared with the sham-operation group(P0.05);the average latency of 3 doses of genstein groups was significantly shortened as compared with the ovariectomized group(P0.05);but no significant difference was found among low,medium and high doses of genstein groups(P0.05).The apoptosis of neurons in the hippocampus of the ovariectomized group was greater than that in the sham ovariectomized group(P0.01).Treatment with 4-week different doses of genstein in the experiment groups significantly decreased the apoptosis of neurons in the hippocampus as compared with the ovariectomized group(P0.05),but no significant difference was noted among the 3 experimental groups and the sham-operation group(P0.05).Conclusions: Genstein can enhance the capabilities of learning and memory of the ovariectomized rats.This improvement may be due to the decrease in the number of apoposis neurons in the hippocampus.
