摘要
采集黑白花奶牛胚胎 2 9枚 ,选用 2 2枚在乙二醇 (EG)、二甲基亚砜 (DMSO) ,添加 4mg/mLBSA的D PBS液 ,按 1∶1∶2的混合溶液 (VS1) ,以及在上述VS1和PBS - [PBS( +) +4mg/mLBSA]的 1∶1混合溶液 (VS2 )中 ,进行玻璃化冷冻 ,将冷冻解冷的 13枚黑白花奶牛胚胎移植于贵州本地受体黄牛 ,妊娠 5头 ,受胎率 38 5 % ( 5 /13) ;产出 3头犊牛 ,产犊率 2 3 1% ( 3/13) ;而采集鼠胚胎 90枚 ,在体积分数为 2 5 %甘油 (G)、体积分数为 5 %丙二醇 (PG)、体积分数为 5 %乙二醇和在体积分数为 2 5 %甘油、体积分数为 2 5 %丙二醇、体积分数 2 0 %FCS PBS混合物溶液 (VS2 )中 ,进行玻璃化冷冻 ,解冻后的鼠胚胎在体外培养至膨胀发育率为 83 3% ( 40 /48) ,移植于受体后 47 5 % ( 19/40 )的胚胎发育成为新生个体。
black and white cow embryos were collected. Among them, selecting 22 effective embryos, through the vitrification of the bovine embryos in a mixture of ethylene glycol, dimethyl sulfoxide, 4 mg/mL BSA D PBS(VS 1) according to 1∶1∶2 and in a mixture of the above VS 1 and PBS-[PBS(+)+4 mg/mL BSA](1∶1)(VS 2),after frozen and thawed, 13 black and white cow embryos had been transplanted to Guizhou local cattles and five cattles had been pregnant. Subsequently three calves had been born. Conception rates were 38 5%(5/13) and birthrates of cattles were 23 1%(3/13). However,collecting 90 mouse embryos, through the vitrification of mouse embryos in a mixture of 25% glycerin, 5% propylone glycol, 5% ethylene glycol(VS 1) and in a mixture of 25% glycerin,25% ethylene glycol and 20% FCS-PBS (VS 2), after frozen and thawed, the developmental rates of expansion in vitre culture were 833% (40/48) and 475%(19/40) of mouse embryos transplanted to recipient mice had grown into newborn individuals.
