摘要
目的观察涤痰汤对帕金森病(PD)药物左旋多巴(LD)毒副反应大鼠纹状体多巴胺转运蛋白(DAT)的影响。方法在PD模型大鼠的基础上腹腔注射左旋多巴/苄丝肼2周制备LD模型,将成功LD模型大鼠随机分为LD模型组、涤痰汤组各9只,另取正常对照组和PD模型对照组大鼠各9只。LD模型组、涤痰汤组分别给予生理盐水、涤痰汤灌胃;PD模型对照组、正常对照组给予生理盐水灌胃(分别为2、4、6周)。每组在2、4、6周各取3只大鼠,行脑纹状体内DAT125I-β-CIT放射自显影,同时用γ-计数器测定DAT放射活性。结果 LD模型组随时间延长DAT数量有减少趋势,且在4、6周时均较PD模型对照组为低;涤痰汤组随干预时间的延长,DAT数量有增加趋势,且在4、6周时均较LD模型组为高,但差异均无统计学意义(P>0.05)。自显影结果与DAT变化基本一致。结论涤痰汤可以缓解帕金森病左旋多巴治疗引起的DAT数量减少。
Objective To observe the effect of Ditan Decoction(Phlegm-dissolving decoction)on dopamine transporter(DAT)in striatum of Parkinson's disease(PD)rats with side-effects by Levodopa(LD).Methods Based on the PD model rats LD models were made through intraperitoneal injection of LD for 2 weeks.The successfully made LD models were randomized into LD model group and Ditan Decoction group,9 rats for each group.And the normal control group and PD model group were set up for comparison,9 rats in each group.The LD model group and Ditan Decoction group were given normal saline and Ditan Decoction respectively by intragastric feeding.PD model group and normal control group were given normal saline by intragastric feeding.In the 2nd,4th,and 6th week,3 rats were taken from each group for the examination of DAT autoradiography in striatum with 125I-β-CIT and the determination of DAT radioactivity with γ-counter.Results With the time going on,the number of DAT of the LD model group had the tendency of decrease,and in the 4th and 6th week,it was lower than in the PD model group.While the number of DAT of the Ditan Decoction group had the tendency of increase,and in the 4th and 6th week,it was higher than in the LD model group.But the difference was not significant(P0.05).The result of autoradiography was basically consistent with it.Conclusion Ditan Decoction might be effective against the decrease of DAT caused by LD treatment of PD.
