摘要
目的探讨复方中药丹星通络汤对大鼠局灶性脑缺血再灌注损伤的保护作用及机制。方法雄性SD大鼠50只,随机分为5组,对照组、模型组、尼莫地平组(尼莫地平10 mg/kg)、方剂小剂量组(复方丹星通络汤7.5g/kg)和方剂大剂量组(复方丹星通络汤15 g/kg),每组10只,连续灌胃5 d。检测血清丙二醛含量、大脑促凋亡基因(Fas)细胞表达,并观察行为学改变。结果模型组大鼠局灶性脑缺血再灌注后血清丙二醛含量、脑组织Fas阳性细胞表达、行为学评分明显高于对照组、尼莫地平组、方剂小剂量组和方剂大剂量组,差异有统计学意义(P<0.05);方剂大剂量组大鼠血清丙二醛含量、脑组织Fas阳性细胞表达、行为学评分较尼莫地平组明显降低,差异有统计学意义(P<0.05)。结论大剂量复方丹星通络汤可有效降低脑缺血再灌注损伤大鼠血清丙二醛含量、Fas阳性细胞表达、行为学评分,对脑缺血再灌注损伤有明显保护作用。
Objective To study the neuroprotection and mechanism of high dose Danxingtongluotang(DX) against cerebral ischemia-reperfusion injury of rats. Methods Fifty SD male rats were divided randomly into 5 groups: control group,model group(ischemia-reperfusion+NS),nimodip- ine group (nimodipine 10 mg/kg, ischemia-reperfusion +NS), small dose DX group (DX 7. 5 g/kg) ,high dose DX group(DX 15 g/kg). There were 10 rats in each group. The 5 groups were fed with nimodipine or DX for consecutive 5 d. The content of MDA in serum was measured, the expression of Fas in brain tissue and behavior scores of rats were examined. Results After cerebral ischemia-reperfusion,the content of MDA in serum, expression of Fas in brain tissue and behavior score of model group increased significantly as compared with other groups (P 〈0.05), the content of MDA in serum,the expression of Fas in brain tissue and behavior scores were decreased significantly in high dose DX group than in nimodipine group and small dose DX group (P 〈 0.05). Conclusion High dose DX has an obvious protection against ischemia-reperfusion injury probably through decreasing the content of MDA in serum,the expression of Fas in brain tissue and behavior scores of rats.
