摘要
目的:观察维拉帕米(Verapamil,Ver)对兔视网膜色素上皮(retinalpigmentepithelium,RPE)细胞增殖的影响,寻找有效、安全、方便的药物以防治增殖性玻璃体视网膜病变(proliferativevitreoretinopathy,PVR)。方法:分别将不同浓度的维拉帕米加入体外培养的兔RPE细胞,用噻唑蓝(MTT)比色法在自动酶标测定仪测540nm处的吸光值A[旧称光密度(OD)],计算出不同浓度条件下的细胞增殖抑制率及50%抑制浓度(fiftypercentinhibitoryconcentration,IC50),同时用2%台盼蓝染色,测定细胞活性。结果:Ver在所设浓度范围内均明显抑制血清诱导的兔RPE细胞的增殖,抑制作用呈剂量依赖性,IC50为16.7659mg/L,超过80mg/L细胞生存受到影响。结论:Ver在较低浓度下即能明显抑制RPE细胞增殖,有希望成为防治PVR的药物之一。
Objective:To investigate the effect of verapamil on surum induced proliferation of rabbit pigment epithelial (RPE) cells and to search for simple and effective medicine on PVR. Methods:The rabbit RPE cells (passage 2 to 5) were cultured with various concentrations of verapamil in DMEM. The rates of inhibition were respectively determined by MTT method, and the cell viability was estimated by trypan blue exclusion test. Results:Verapamil at all given concentrations significantly inhibited the serum induced proliferation of RPE cells. At concentrations of 80 mg/L and lower, it had no effect on cell viability. The IC 50 of verapamil was 16.7659 mg/L.Conclusion:Verapamil significantly inhibits RPE cell promising drug on PVR.
