砂仁及其混淆品的ITS2序列鉴定 被引量:25

摘要 目的应用内转录第二间隔区(ITS2)序列及其二级结构对砂仁混淆品进行鉴定。方法应用CLUSTALX1.83软件进行序列比对,比对后的长度为252bp。用MEGA4.1计算种间、种内遗传距离,并构建系统进化树。结果 ITS2序列可以将砂仁的三个来源阳春砂Amomum villosum、绿壳砂Amomum villosum var.xanthioides或海南砂Amomum longiligulare互相区分开,并与伪品艳山姜Alpinia zerumbet、山姜Alpinia japonica、草果Amomum tsao-ko及同属其它近缘种区分开。结论推荐ITS2作为砂仁及其伪品鉴定用条形码序列。 Objective Identification of Amomi fructus and its adulterants based on ITS2 primary sequence and secondary structure.Method CLUSTALX 1.83 software was used to align the sequences and the length of the aligned sequence was 252 bp.The MEGA4.1 software was used to calculate the K-2P distance between intra-and inter-specific species.The NJ tree was also been built.Result The ITS2 sequence would be used to identify Amomum villosum,Amomum villosum var.xanthioides,Amomum longiligulare and its adulterants,such as Alpinia zerumbet,Alpinia japonica,Amomum tsao-ko etc.Conclusion ITS2 sequence was recommended as barcode to identify Amomi fructus and its adulterants.
