摘要
稻瘟菌是危害水稻生产的世界性病原菌,也是研究植物与病原菌互作系统的重要模式生物。镰孢菌是危害小麦等重要农作物的主要病原菌,且能产生真菌毒素而对农产品造成污染。随着稻瘟菌和镰孢菌全基因组测序成功,如何利用基因组序列来鉴定和分析基因功能成为当前主要挑战。反向遗传学方法为进一步研究稻瘟菌基因功能提供了重要基础。综述了基因敲除、插入突变、基因沉默以及定向诱导基因组局部突变等一些热点反向遗传学方法及其在稻瘟菌和镰孢菌功能基因组分析中的应用进展。
Magnaporthe grisea is a worldwide pathogenic bacterium that endangers rice production,and it is also an important model organism for studying interactive system between plants and pathogenic bacteria.Fusarium is a major pathogenic bacterium that endangers important crops like wheat,and it can produce fungal toxin to pollute agricultural products.With the success of genome sequencing for Magnaporthe grisea and Fusarium,how to use genome sequencing for identify and analyzing gene function has became the major challenge at present.Meanwhile,reverse genetics method provides important basis for further study on gene functions of Magnaporthe grisea.In this paper,some hotspots of reverse genetics method,such as gene knockout,insertion mutation,gene silencing and directing inducing local mutation in genome,were introduced,and the applications of these hotspots in analyzing functional genome of Magnaporthe grisea and Fusarium were also summarized.
