功能前伸大鼠下颌翼外肌胰岛素含量昼夜节律的研究

摘要 目的通过研究功能矫治器前伸大鼠下颌,胰岛素在翼外肌局部含量昼夜波动规律以及在不同时段戴用同一矫治器时此内在节律的变化特征,探讨功能矫治器最佳戴用时间以及戴用时间长短与效应的关系。方法 54只四周龄雄性wistar大鼠随机分成3组,使用自制咬合前伸矫治器,取大鼠两侧翼外肌用放射免疫法计测翼外肌内胰岛素含量。应用时间生物学统计方法-余弦法处理全部计测值判断是否合乎节律特征。结果各组胰岛素含量拟合余弦曲线分别为:对照组胰岛素的含量拟合余弦曲线方程为Yi=32.1+5.3cos(15 ti+177)。12 h加力组胰岛素的含量拟合余弦曲线方程为Yi=37.1+6cos(15 ti+185)。24 h加力组胰岛素的含量拟合余弦曲线方程为Yi=39.6+5.7cos(15 ti+188)。3组皆具有生物节律性,且24 h加力组胰岛素的含量高于12 h加力组,12 h加力组高于对组照胰,差异具有统计学意义。结论大鼠翼外肌的生长改建,昼夜并非均匀一致,而是表现出有规律的昼夜波动过程,即白天增生改建较强,夜晚较弱。戴用功能矫治后生长加速,白天12 h戴用功能矫治器即可产生矫治效果。戴用时间延长会增加功能矫治器的矫治效应。 Objective In order to determine which time of the day is best to wear functional appliances and if It is the longer the better to wear functional appliances, the author investigates the circadian rhythm of the level of endogenous Insulin in growing rat condylar cartilages and the changes after functional mandibular protrusion in daytime (8:00 -20:00) or all the day (00:00 -24:00). Methods 54 4 -week -old Wistar rats were divided equally and randomly into 3 groups, the rats in the 12 - hour wearing group were put on the functional appliances in daytime (8:00 - 20:00) and the 24 - hour wearing group were put on the functional appliances all the day (00:00 -24:00). After 7 days, all rats were sacrificed at 0:00, 4:00, 8:00, 12:00, 16:00, 20:00 in a day. Endogenous Insulin level in lateral pterygoid muscles were measured by using radioimmunoassy. Dates were processed using macroscopic and microscopic analysis. Results Fitting equations of the control group is Yi = 32. 1 + 5.3cos( 15 ti + 177) (P 〈 0. 05 ). fittingquation and characteristic values of the 12 - hour wearing group is Yi = 37. 1 + 6cos ( 15 ti + 185 ) ( P 〈 0. 05 ). Fitting equation and characteristic values of the 24 - hour wearing group is Yi = 39.6 + 5.7cos ( 15 ti + 185 ) ( P 〈 0. 05 ). Conclusion The growth of rat lateral pterygoid muscle alterations, day and night is not uniform, but showed a regular diurnal fluctuations in the process of rebuilding a strong proliferation of the day, the night is weak. After functional appliance wear the growth of lateral pterygoid muscle is accelerated during the day and 12 hours to produce treatment results.
