HIF-1α在缺氧性心搏骤停大鼠的表达和变化研究

摘要 目的探讨缺氧诱导因子-1α(HIF-1α)在缺氧性心搏骤停大鼠肺脏和脑组织的表达变化。方法将45只雄性SD大鼠随机分为3组,分别为CO中毒心搏骤停组(CO中毒组)、气管夹闭窒息性心搏骤停组(气管夹闭组)和对照组。大鼠心搏骤停模型制备成功后,免疫荧光和ELISA检测HIF-1α在大鼠心搏骤停15 min时血浆、肺和脑组织中的表达和变化情况。结果 CO中毒组及气管夹闭组血浆HIF-1α水平明显增加,且显著高于对照组(P<0.01),其中CO中毒组明显高于气管夹闭组(P<0.05)。CO中毒组及气管夹闭组肺组织HIF-1α表达明显高于对照组(P<0.05),其中CO中毒组高于气管夹闭组(P<0.05);CO中毒组大脑皮层HIF-1α表达明显高于对照组和气管夹闭组(P<0.05),但气管夹闭组与对照组差异无统计学意义(P>0.05)。结论 HIF-1α在缺氧性心搏骤停早期表达明显增强,其表达强度与缺氧程度、快慢速度等相关,可作为评价心搏骤停预后的参照指标。 Objective To investigate the expression of hypoxic-inducible factor-1α(HIF-1α) in rats with hypoxic cardiac arrest.MethodsForty-five male Sprague Dawley(SD) rats were divided randomly into three groups,CO group in which hypoxic cardiac arrest caused by CO poisoning,occlusion group in which hypoxic cardiac arrest caused by tracheal occlusion and the normal control group.The samples of blood,lung and brain were taken after 15 min when hypoxic cardiac arrest rats models were duplicated,the expression of HIF-1α was measured by immunofluorescence and ELISA.ResultsThe levels of HIF-1α in plasma were significantly higher in both hypoxic cardiac arrest groups than that in control group(P0.01),and it was higher in CO group than that in occlusion group(P0.05).The expression of HIF-1α was increased significantly in experimental group than that in control group(P0.05) both in lung and brain,and it was also higher in CO group than that in occlusion group in the lung tissue(P0.05),but there was no difference between occlusion group and control group in brain(P0.05).ConclusionThe expression of HIF-1α is increased in the early stage in rats with hypoxic cardiac arrest,and the intensity of expression is related with the severity and speed of hypoxic,so it is a valuable marker to predict prognosis of rats with hypoxic cardiac arrest.
