摘要
目的 建立谷氨酸诱导的大鼠星形胶质细胞肿胀模型,观察 Dyn A1 ~13 对谷氨酸诱导的大鼠星形胶质细胞肿胀的影响。方法 [3 H] 3氧甲基 D葡萄糖摄取测定细胞含水量。结果 ①给予05 ,10 ,100 m mol· L- 1 的谷氨酸作用1 h 均可引起细胞的含水量增加。②强啡肽 A( Dyn A) 可降低谷氨酸诱导肿胀的大鼠星形胶质细胞含水量。③κ受体拮抗剂nor B N I可阻断 Dyn A1 ~13 降低谷氨酸诱导的肿胀大鼠星形胶质细胞含水量的作用。结论 谷氨酸可诱导大鼠星形胶质细胞肿胀; Dyn A1 ~13
AIM To establish a medol of astrocyte swelling induced by glutamate and to study the effect of dynorphin A 1~13 on astrocyte swelling. METHODS 3 O methyl D glucose uptaking was used to determine water content of cells. RESULTS ①Glutamate(0 5, 1 0, 10 0 mmol·L -1 ) increased the water content of astrocyte at an hour. ② Dynorphin A 1~13 could significantly decrease the water content of swelling astrocytes induced by glutamate. ③nor BNI, a κ opioid receptor antagonist, could inhibit the effect of dynophin A 1-13 on water content of swelling astrocytes. CONCLUSION Glutamate can induce the swelling of astrocyte. Dynorphin A 1~13 may reduce astrocytes swelling induced by glutamate via κ receptor.
