摘要
人脐静脉内皮细胞体外培养对于血管内皮细胞的结构和功能研究提供了一条十分有效的途径。本文研究了影响人脐静脉内皮细胞体外培养的几个因素。实验提示:存活细胞数随脐带离体时间而减少,脐带离体24h 尚有50%的存活细胞;培养液中血清的最适浓度为10～16%;胶原和纤维结合蛋白等细胞外基质成分均有利于内皮细胞粘附和生长;肝素。
Culture of human umbilical-cord vein endothelial cell in vitro provides a conven-ient way to study the structure and function of human endothelial cell.In this rese-arch,we studied some factors which effect the propagation of human umbilical-cordvein endothelial cell in vitro.The results show:the longer umbilical-cords are deli-vered from body,the less the alive endothelial cells exist;alive cells are about fiftypercent when umbilical-cords have been delivered from body for twenty four hours,the best serum conceentration in culture medium is ten to fifteen percent;The com-ponents of extracellular matrix,such as collagen and fibronectin can promote endo-thelial cells adhension and propagation in vitro;Heparin or theombin,without usedwith ECGF,has no notable effects on human endothelial cells propagation in vitro.
