狂犬病病毒SRV-9株对小鼠脑神经原代细胞的感染及毒力观察 被引量:1

Observation on infection and virulence of rabies virus SRV-9 strain on murine primary neuron cells

摘要 为建立一种狂犬病病毒(RV)感染原代神经细胞的体外培养方法,分离培养了新生小鼠脑神经原代细胞,并进行了RV感染试验,利用免疫荧光技术和免疫细胞化学试验检测了神经细胞中的RV及其对被感染细胞的影响。结果显示,RV可感染分离的原代神经细胞,并可以检测到针对RV的特异性荧光。表明,建立了可培养RV的原代神经细胞的制备方法,并且能够观察到原代神经细胞适应RV感染时的毒力变化。 To establish a method used for infecting primary neuron cells by rabies virus in vitro and made some observations on toxicity and adaptability after infection,primary neuron cells were isolated and cultured from newborn mice which were infected with rabies virus SRV-9 strain later,and the virus was detected and the influence of the virus on the primary neuron cells was analyzed by the immune fluorescence technology and the immunocytochemistry tests.In result,the murine primary neuron cells were successfully infected by rabies virus and the specific fluorescence was detected.These results showed that the method for preparation of primary neuron cells suitable for culturing rabies virus was successfully developed and its toxicity change can be observed when murine primary neuron cells adapt to the infection of the virus.
