大豆不同外植体组织培养及再生研究 被引量:22

摘要 利用不同大豆品种的顶芽、胚轴、子叶节、子叶、叶片等做为外植体,进行组织培养再生研究。结果表明:不同大豆品种诱导分化芽的能力差别较大,供试品种中,辽豆11号和辽豆10号的再生能力较强,诱芽率分别为39.8%和36.0%。对诱芽率的影响,经L_(16)(4)~5正交实验可知外植体>品种>培养基。外值体对诱芽率影响依次为:子叶节>顶芽>子叶>下胚轴。6-BA对不同外植体诱导芽发生的适宜浓度不同,顶芽、下胚轴为0.5mg·l^(-1),子叶为1.0mg·1^(-1),成熟子叶为4.0mg·L^(-1)。 The regeneration of tissue culture of soybean (Glycin max L.) was Studied from terminal bud, hypocotyl,cotyledonary node and cotyledon in several varities of soybean. The result indicates that the difference of bud initiation was obvious on varieties of soybean. Among varieties supplied, Liaodou 11. and Liaodou 10 had comparatively high frequency of bud-inducing, with 39.8% and 36% respectively. According to the L_(16) (4)~5 orthogonal test, teh affecting,order on the rate of bud-inducing were exophyte, genotypes and culture media, the affecting order of various exophytes were cotyledonary node, terminal bud and cotyledon. Bud-inducing of exophytes needs different concentitation of 6-BA, terminal bud and hypocoty for 0.5mg·L^(-1), cotyledonary node 1.0mg·L^(-1) and cotyledon for 4.0mg·L^(-1).
