利用胚珠培养技术培育棉属种间杂种植株的研究 被引量:2

STUDIES IN GETTING HYBRIDS PLANTS OF INTERSPECIFIC\=HYBRIDIZATION THROUGH OVULE CULTURE IN VITRO

摘要 本研究利用陆地棉×中棉、陆地棉×海岛棉、独秆突变体×海岛棉、海岛棉×中棉杂交,进行胚珠培养,获得种间杂种植株。结果表明:这4个杂交组合均获得一定频率的胚(包括成熟胚和不成熟胚)及成熟胚萌发的苗;同一组合的杂种胚珠,在不同培养基上获得的胚和成熟胚的频率不同;不同组合的杂交胚珠,在同一培养基上培养的结果有明显差异;进行杂种胚珠培养的适宜时期为授粉后4~6d,2d的胚珠培养易产生愈伤组织,成胚率低,8d后幼胚败育或落桃;适合杂种胚珠早期生理条件的培养基为:BT基本培养基+NH4Cl1000mg/l+IAA15mg/L+KT02mg/L+GA006mg/l;氨态氮能提高成胚率;单一的培养基不能满足杂种胚珠的培养需要。 Ovule culture was made on Gossypium hirsutumG.arboreum, G.hirsutumG.barbadense, G.vc singal stem mutantG.barbadense, G.barbadenseG.arboreum. After 8590 days of culture embryos (including matured and unmatured ones) as well as seedling from matured embryo were obtained from these 4 crosses with a certain frequency. The results of experiment showed that the differences were significant between embryo frequency and germination percentage of embryos with ovule culture on different media and the culture with ovules from different crosses, and on the same medium the difference was great.\;The ovules from hybrids having developed for different days after pollination were cultured on BT,BT medium in the darkness. Embryo development with a high frequency was obtained with ovules from G.hirsutumG. barbadense and G.hirsutumG. arboreum 46 days after pollination.
