八肽胆囊收缩素对新生小鼠大脑皮层神经元生长的影响 被引量:6

EFFECT OF OCTAPEPTIDE CHOLECYSTOKININ ON THE GROWTH OF CORTICAL NEURONS OF NATAL MICE

摘要 目的:探讨八肽胆囊收缩素(octa-peptide cholecystokinin,CCK8)对体外培养的新生小鼠大脑皮层神经元生长的影响。方法:原代培养新生小鼠大脑皮层神经元,采用烯醇化酶(neuron-specific enolase,NSE)免疫细胞化学染色法鉴定皮层神经元,培养24h的皮层神经元随机分为对照组和CCK8刺激组(终浓度为10-7mol/L),作用24h、48h、72h、96h后,倒置相差显微镜下行动态观察,MTT法检测细胞的生长活性并以OD值绘制生长曲线。结果:CCK8处理后,有突起的神经元数目增加,存活时间延长,同一时间点的细胞活性明显增强,差异有统计学意义(P<0.05)。结论:CCK8能促进皮层神经元的生长,延长其存活时间。 Objective:To explore the effects of octapeptide cholecystokinin(CCK8)on the growth of the cerebral cortical neurons from new born mice.Methods:Cultured neuron was identified by neurone specific enolase(NSE) immunohistochemical staining.After cultured for 24 hours,neurons were divided into CCK8 stimulation group and control group at random.After both groups were stimulated with DMEM and CCK8(10-7 mol/L) respectively for 24,48,72 and 96 hour,the morphology of neuron was observed by the inverted light microscope and cell viability was analyzed by MTT assay.Results:The number of neurons with neurite and the cell viability in CCK8 group increased compared with the control group(P〈0.05).CCK8 increased the survival of neurons.Conclusion:CCK8 could improve the growth of the cortical neurons and enhance the survival time.
