天然化合物TTS-12对白念珠菌细胞凋亡的影响 被引量:3

摘要 目的研究天然化合物TTS-12(Tribulus terrestris S-12)对白念珠菌细胞凋亡的影响。方法应用透射电镜技术观察TTS-12作用于白念珠菌后细胞超微结构的变化,采用Annexin V及PI测定TTS-12对白念珠菌细胞凋亡率的影响,采用POLARstar多功能微板检测仪通过荧光强度检测TTS-12对白念珠菌细胞内活性氧(reactive oxygen species,ROS)水平的影响。结果与空白对照组相比,TTS-12可明显诱导白念珠菌细胞凋亡;8μg/ml的TTS-12可诱导58%的细胞凋亡;TTS-12对白念珠菌细胞内ROS产生的促进作用具有剂量依赖性,并随着时间的延长对ROS产生的促进作用也随之增强。结论 TTS-12可诱导白念珠菌细胞凋亡,其机制可能与升高白念珠菌细胞内ROS水平有关。 Objective To study the effect of the natural compound TFS - 12 on apoptosis of Candida albicans. Methods The morphological changes in the cells treated with TFS - 12 were shown by transmission electron micrography. The apopto- sis rate was determined using annexin V and PI. Reactive oxygen species (ROS) was detected on the POLAR star Galaxy. Results TTS - 12 significantly induced C. albicans apoptosis, compared with the control group. The apoptosis rate of C. albicans was 58% at TTS- 12 concentrations of 8 μg/ml. Compared with normal cells, the apoptotic cells induced by TTS -12 showed increased endogenous ROS generation in a dose - dependent and time - dependent manner. Conclusion TTS - 12 induces Candida albicans apoptosis by enhancing endogenous ROS generation.
