摘要
目的:研究强心饮对肿瘤坏死因子α(tumor necrosis factorα,TNF-α)诱导新生大鼠心肌成纤维细胞(cardi-ac fibroblasts CFs)增殖和胶原合成的影响,探讨其抑制心肌纤维化的作用机制。方法:采用胰酶消化法分离培养乳鼠CFs,建立TNF-α诱导新生大鼠CFs纤维化病理模型,分别采用四氮唑蓝(MTT)比色法、羟脯氨酸法检测不同浓度强心饮含药血清对CFs增殖和胶原含量影响。结果:TNF-α能够刺激CFs增殖和胶原合成,与空白组相比差异显著(P<0.05);强心饮抑制TNF-α诱导CFs增殖和胶原合成(P均<0.05)。结论:强心饮通过抑制TNF-α诱导的CFs增殖和胶原合成,发挥有效的抗心肌纤维化作用。
Objective:To investigate the effect of Qiangxin Decoction on the proliferation and collagen synthesis in neonatal rats cardiac fibroblast(CFs) induced by tumor necrosis factor α(TNF-α),and to explore the mechanism of Qiangxin Decoction on reversing cardiac fibrosis.Methods:Cardiac fibroblasts derived from neonatal rats were cultured with enzymatic dissociation,and CFs pathological model was induced by tumor necrosis factor α(TNF-α).Thiazolyl blue MTT assay and hydroxyproline were employed to measure Cfs proliferation and collagen synthesis in order to determine the effect of different dosage decoction on CFs proliferation and collagen synthesis.Results:Compare with control group,TNF-α could induce cardiac fibroblast proliferation and increase collagen synthesis significantly(P〈0.05),while Qiangxin Decoction inhibit Cfs proliferation and collagen synthesis(P〈0.05).Conclusion:Qiangxin Decoction could reverse cardiac fibrosis,which may be related to inhibit CFs proliferation and collage synthesis induced by TNF-α.
