摘要
建立了HPLC法测定兔血浆中AF-2364的浓度,血浆样品经蛋白沉淀后以正己烷-乙酸乙酯(1:4)萃取,采用C_(18)色谱柱,以0.1%三氟乙酸溶液-乙腈(55:45)为流动相,以卡马西平为内标,检测波长302 nm。结果AF-2364在0.1~20μg/ml浓度范围内线性关系良好,日内和日间RSD为2.1%~11.2%,提取回收率和方法回收率约77.51%和108 89%。
An HPLC method was established for the determination of AF-2364 in rabbits plasma.Samples were extracted by n-hexane-ethyl acetate(1:4) after protein precipitation.A C_(18) column was used with the mobile phase of 0.1%trifluoroacetic acid solution-acetonitrile(55:45) at the detection wavelength of 302 nm.Carbamazepine was used as the internal standard.The calibration curve was linear in the concentration range of 0.1 - 20μg/ml.The inter- and intra-day RSDs were 2.1%- 11.2%.The extraction and method recoveries were about 77.51%and 108.89%.
