摘要
利用从北美鹅掌楸(Liriodendron tulipifera Linn.)EST序列中开发的176对SSR引物,以国内木兰科10属43种的DNA样品为材料,检测了该批引物在木兰科各属间的通用性。132对引物在北美鹅掌楸中有效扩增,108对在鹅掌楸(L.chinense Sarg.)中有扩增产物,在鹅掌楸属内的通用率为81.8%;在含笑属(Michelia)、木莲属(Manglietia)、木兰属(Magnolia)、合果木属(Paramichelia Hu)、观光木属(Tsoongiodendron)、焕镛木属(Woony-oungia Law)、拟单性木兰属(Parakmeria Hu)、盖裂木属(Talauma Juss)、华盖木属(Manglietiastrum Law)中通用率则分别为24%~71%、37%~47%、27%~41%、49%、44%、36%、31%、29%、24%。利用北美鹅掌楸、鹅掌楸、玉兰(Magnolia denudata)和云南拟单性木兰(Parakmeria yunnannensis)各6个基因型检测了该批引物的扩增多态性,其多态性引物比率分别为55%、45%、48%和39%。最后,讨论了该批EST-SSR引物在木兰科植物相关研究中的潜在应用前景。
An experiment was conducted to study the cross-genus transferability of 176 pairs of EST-SSR primers developed from Liriodendron tulipifera Linn.and amplified in 43 species from 10 genera of Magnoliaceae in China.Among 176 pairs of SSR primers,132 pairs of primers were successfully amplified in L.tulipifera,and 108 pairs had amplification products in Liriodendron chinense Sarg.The transferability within the genus Liriodendron was 61%.The cross-genus transferability of Michelia,Manglietia,Magnolia,Paramichelia,Tsoongiodendron,Woonyoungia,Parakmeria,Talauma and Manglietiastrum was 24%-71%,37%47%,27%-41%,49%,44%,36%,31%,29% and 24%,respectively.Furthermore,the amplification polymorphisms of these primers in L.tulipifera,L.chinense,Magnolia denudate,and Parakmeria yunnannensis were also detected,and the ratios of polymorphic primers were 55%,45%,48% and 39% respectively.The potential applications of these EST-SSR primers in Magnoliaceae were also discussed.
