染料木素和青藤碱对佐剂型关节炎大鼠血清IL-17、Tracp的影响 被引量:7

Effects of Genistein and Sinomenine on Serum IL-17 and Tracp in Adjuvant-Induced Arthritis Rats

摘要 目的:研究染料木素和青藤碱对佐剂型关节炎(AA)大鼠血清白介素17(IL-17)、抗酒石酸酸性磷酸酶(Tracp)含量的影响,以初步探讨其对AA大鼠骨免疫系统的调节作用。方法:通过给大鼠左后足跖皮下注射完全弗氏佐剂建立AA大鼠模型。注射诱导后第2天大鼠出现急性关节炎症状,踝关节不同程度肿胀,炎症持续第21天开始给予药物干预。染料木素组给予染料木素(1 mg/200 g.d-1)灌胃,青藤碱组给予青藤碱(3 mg/200 g.d-1)灌胃,配伍组给予染料木素(1 mg/200 g.d-1)和青藤碱(3 mg/200 g.d-1)灌胃,甲氨喋呤组给予甲氨喋呤(1 mg/200 g.w-1)灌胃。连续干预40d后用酶联免疫吸附法测定大鼠血清IL-17和Tracp的含量。结果:①配伍组与青藤碱组和甲氨喋呤组比较,IL-17、Tracp含量均明显降低(P<0.05,P<0.01);与染料木素组比较,IL-17、Tracp含量下调但差异不显著(P>0.05)。②染料木素组Tracp含量明显低于青藤碱组和甲氨喋呤组(P<0.05);染料木素组IL-17含量低于青藤碱组、高于甲氨喋呤组,但差异无统计学意义(P>0.05)。结论:染料木素和青藤碱合用能发挥协同作用,降低AA大鼠血清IL-17、Tracp含量。 Objective:To study the effects of genistein and sinomenine on serum interleukin 17(IL-17) and tartrate-resistant acid phosphatase(Tracp) in adjuvant-induced arthritis(AA) rats,and further investigate their regulative action on osteoimmunology system.Methods: Complete Fruez adjuvant was hypodermicly injected into the left rear paw of the rats to establish AA model.Acute arthritis in rats induced on the day after injection displayed different degrees of ankle swelling,and the drugs were administered since inflammation continued to the 21st day.Genistein group was fed with genistein(1mg/200g·d),sinomenine group with sinomenine(3mg/200g·d),compatibility group with both genistein(1mg/200g·d) and sinomenine(3mg/200g·d),and methotrexate group with methotrexate(1mg/200g·w).The contents of serum IL-17 and Tracp were detected by enzyme-linked immunosorbent assay after intervention for 40 days.Results: ①Compared with sinomenine group and methotrexate group,the contents of IL-17 and Tracp were significantly lower in compatibility group(P0.05,P0.01);there was no significant difference in the contents of IL-17 and Tracp between compatibility group and genistein group(P0.05).②The content of Tracp was obviously lower in the genistein group than in the sinomenine group and methotrexate group(P0.05);the content of IL-17 was similar in the genistein group in comparison with the sinomenine group and the methotrexate group(P0.05).Conclusion: Combined use of genistein and sinomenine can reduce the contents of IL-17 and Tracp in the adjuvant-induced arthritis rats.
