SDF-1/CXCR4生物学轴在乳腺癌中的表达及其临床意义 被引量:4

摘要 目的检测趋化因子基质细胞衍生因子1(SDF-1)及其受体CXCR4在乳腺浸润癌、导管原位癌及癌旁组织中的表达,分析两者与乳腺浸润癌临床病理学特征的关系,探讨SDF-1/CXCR4生物学轴在乳腺癌发生发展过程中的的作用。方法采用免疫组化方法检测80例乳腺浸润癌、15例导管原位癌和15例癌旁组织中SDF-1及CXCR4蛋白的表达。应用RT-PCR检测40例乳腺浸润癌、5例导管原位癌和5例癌旁组织中SDF-1及CXCR4 mRNA的表达。结果免疫组化检测结果显示,SDF-1、CXCR4在乳腺浸润癌与导管原位癌,导管原位癌与癌旁组织中的表达差异均有统计学意义(P<0.05),其中CXCR4表达与乳腺癌的淋巴结转移和分期有关(P<0.05)。RT-PCR检测结果显示,SDF-1与CXCR4 mRNA在乳腺浸润癌与导管原位癌,导管原位癌与癌旁组织中的表达差异均有统计学意义(P<0.05)。SDF-1、CXCR4蛋白及mRNA在乳腺浸润癌中的表达呈正相关(P<0.05)。结论 SDF-1/CXCR4生物学轴在促进乳腺癌的发生发展及侵袭转移过程中起促进作用。 Objective To analyze the clinicopathological significance of ligand SDF-1 and its receptor CXCR4 expression in breast carcinoma,ductal carcinoma in situ and adjacent tissues and clarify the roles of SDF-1/CXCR4 biological axis in the pathogenesis of breast cancer.Methods SDF-1 and CXCR4 protein expression were examined in 80 cases of breast carcinoma,15 cases of ductal carcinoma in situ and 15 cases of adjacent tissue by immunohistochemistry.SDF-1 and CXCR4 mRNA expression were determined in 40 cases of breast invasive carcinoma,5 cases of ductal carcinoma in situ and 5 cases of normal tissue adjacent to cancer tissues by RT-PCR.Results The protein and mRNA expressions of SDF-1 and CXCR4 were different in breast carcinoma and ductal carcinoma in situ,ductal carcinoma in situ and adjacent tissues(P 〈 0.05).The expression of CXCR4 protein was related to lymph node metastasis and breast cancer stage(P 〈 0.05).Both protein and mRNA expression of SDF-1 and CXCR4 were positively correlated in breast carcinoma(P 〈 0.05).Conclusion SDF-1/CXCR4 biological axis might play an important role in promoting breast cancer growth and metastasis.
