摘要
目的建立高效液相色谱-质谱联用测定人血浆中红霉素胺浓度的方法。方法以卡马西平为内标,血浆经碱化采用二氯甲烷提取,色谱柱为Hypersil C18柱(2.1 mm×150 mm,5μm);流动相为0.1 mol.L-1醋酸铵溶液-乙腈-甲醇(65∶30∶10)(醋酸调pH 6.0)。结果红霉素胺血浆浓度在3.1~736 ng.mL-1与峰面积有良好线性关系,方法回收率>89%,日内、日间RSD均<10%。结论该方法灵敏、准确,可用于地红霉素的药动学研究。
Objective To establish a sensitive liquid chromatographic-electrospray ionization mass spectrometric method for the determination of the active metabolite of dirithromycin,erythromycylamine.Methods We mixed the plasma with some solutions.After the centrifugation,N2 dried and dissolved in the mobile phase,samples were separated by a Hypersil C18 reversed-column(2.1 mm×150 mm,5 μm),with the mobile phase consisting of 0.1 mol·L-1 ammonium acetate-acetonitrile-methanol at 65∶30∶10(v/v/v) and pH was adjusted to 6.0 by acetic acid.Carbamazepin was used as the internal standard.Results The method demonstrated linearity 3.1-736 ng·mL-1(r=0.996 2).The recovery of the method was higher than 89%.Intra-and inter-day RSDs were less than 10%.Conclusion The method is sensitive,fast and accurate,which can be used in the monitoring of therapeutic drugs and human pharmocokinetic study of dirithromycin.
