胰岛素和胚胎培养密度对猪孤雌胚早期体外发育的影响 被引量:3

摘要 为了探讨胰岛素和胚胎培养密度对猪孤雌胚早期体外发育的影响,进一步优化猪早期胚胎培养体系,分别进行了胚胎培养液(NCSU-23)中添加不同批号和浓度的胰岛素以及不同胚胎培养密度(培养滴体积)对猪孤雌胚早期胚胎体外发育的影响试验,并于第2、7天分别统计胚胎分裂率和囊胚发育率。结果表明:①在NCSU-23中添加5.0μg/mL批号为I-1882(Sigma公司)的胰岛素,其囊胚发育率最高,为(33.48±12.79)%;②在体外培养25枚猪孤雌胚时,按胚胎数(枚)l培养液体积(μl)为1∶2.0~1∶3.0的比例进行培养,其培养效果较好。 The objective of this study was to evaluate the effect of insulin and culture density on the development of porcine parthenogenetic embryo in vitro and optimize the culture system of porcine early embryo.The porcine parthenogenetic embryos were cultured in NCSU-23 medium supplemented with different concentrations of insulin.Further,25 embryos per group were randomly cultured in culture medium with the proportion(1∶5,1∶3,1∶2,1∶1.5,1∶0.5) of embryo to culture volume(μl).The rates of cleavage and blastocyst formation were recorded on 2nd,7th day,respectively.The results showed that the formation rate of blastocyst in parthenogenetic embryo was the highest(33.48±12.79 %) as adding 5 μg/mL of insulin to NCSU-23.The blastocyte rate of parthenogenetic embryo was the highest in the embryo density of 1∶2.0-1∶3.0,the ratios of embryo to volume being 1∶2-1∶3 were beneficial to the in vitro development of porcine parthenogenetic embryo.
