摘要
以重组NDV HN 基因禽痘病毒(rFPV)为研究对象,观察重组病毒在鸡胚成纤维细胞上表达的动态变化,并对表达蛋白生物学活性进行了检测。结果表明:重组病毒感染鸡胚成纤维细胞12 h 即可检测到表达蛋白,72 h 左右表达量明显增多;利用Western blot可检测到68 kD和74 kD两种分子量的蛋白,与NDV HN蛋白糖基化和非糖基化蛋白形式的分子量一致。表达蛋白具有红细胞吸附活性和神经氨酸酶活性,该活性可被NDV HN单抗所抑制。由此进一步证明重组NDV HN表达蛋白具有与天然NDV HN 蛋白相似的生物学活性,可作为研制NDV 生物工程疫苗目的基因。
By im m unofluorescence test and Western blot analysis,it w as found that chicken em bryo cells transfected w ith recom binant virus expressed tw o kinds of HN sized proteins.(M.W.74 kD;M.W.68 kD).Both of them w ere HN specific proteins and could react w ith anti NDV HN antibody. Their m olecular w eights coincided w ith those of glycosylated and unglycosylated NDVHN proteins.Cells transfected w ith recom binantrFPV could absorb chicken red blood cells(RBC),and the cells surfaces exhibited neuram inidase activity. Both of these activities could be inhibited by anti NDV HN antibodies. The experim ent results show that the recom binant containing NDV HN gene can serve as a candidate for biologicalvaccine ofNDV.
