摘要
采用热抽提和TritonX100 处理2 种方法提取大肠杆菌EL1 株和肺炎克雷伯氏杆菌EL2 株表面抗原,经胃蛋白酶处理、SDSPAGE 及免疫印迹分析表明,EL1 、EL2 株主要表面抗原耐热,对胃蛋白酶不敏感,分子量分别是20KD、33KD 和36KD、43KD,其抗原成分是多糖或多糖磷脂和蛋白质的复合物。二菌株表面抗原具有各自菌种的特征,可作为菌种分类鉴定的重要指标。此外,热抽提法提取细菌表面抗原的效果优于Triton X100。
The surface antigens of E coli(Strain EL 1) and KL Pneumoniae (Strain EL 2)were extracted by heat treatment or by triton X 100 treatment and were treated with proteiase k The antigens,after analysis by polyacryamide gel electrophoresis and immunoblotting,have been shown to be resistant to heat and nonsensitive to proteiase k The molecular weights of the antigens were 20KD、33KD and 36KD、43KD respectively They were composed of polysaccharid or the complex of polysaccharid phosphatied and protein Because the surface antigens in two strains were not common,they had the species character and could be considered a key index in classified identification of species Moreover,the effect of heat extraction in extracted resistance heat surface antigen of bacteria was better than Triton X 100
