摘要
目的：探讨 Ｓ Ｖ４０ 早期基因编码产物大 Ｔ 抗原（ Ｔａｇ） 表达及与抑癌蛋白ｐ Ｒｂ 的相互作用在人脑肿瘤发生发展中的意义。方法：采用免疫共沉淀及 Ｗｅｓｔｅｒｎ 印迹法检测４３ 例人脑肿瘤组织及５ 例正常人脑组织中 Ｔａｇ 的表达，并对１５ 例 Ｔａｇ 阳性瘤组织检测 Ｔａｇ － ｐ Ｒｂ 复合物的存在。结果： Ｔａｇ 在５ 例室管膜瘤及２ 例脉络丛乳头状瘤中全部表达，垂体腺瘤（５／６） 、星形胶质细胞瘤（７／１０） 、脑膜瘤（４／６） 、多形性胶质母细胞瘤（２／５） 及髓母细胞瘤（１／５） 均有 Ｔａｇ 的表达； ３ 例少枝胶质细胞瘤、１ 例松果体瘤及５ 例正常人脑组织无 Ｔａｇ 表达。１５ 例 Ｔａｇ 阳性瘤组织中均发现 Ｔａｇ 与ｐ Ｒｂ 形成特异性复合物。结论：在人脑肿瘤组织中 Ｔａｇ 广泛表达， Ｔａｇ 可与ｐ Ｒｂ 形成特异性复合物， Ｔａｇｐ Ｒｂ特异性复合物的形成导致ｐ Ｒｂ 失活，可能是人脑肿瘤发生发展的一个重要机理。
Objective:To research the role of SV40 early region gene coding product large tumor antigen (Tag) and the interaction between Tag and tumor suppressor pRb in human brain tumorigenesis Methods:Tag was investigated by immunoprecipitation and Western blot in 43 cases of human brain tumors and 5 cases of normal brain tissues.Tag pRb specific complex was screened in 15 Tag positive cases.Results:Tag was found in all of 5 ependymomas and 2 choriod plexus papillomas, as well as in pituitary adenomas (5/6), astrocytomas (7/10),meningiomas (4/6),glioblastomas multiforme (2/5),medulloblastomas (1/5).None of the 5 normal brain tissues was positive for Tag,nor were 3 oligodendrogliomas and 1 pineocytoma. Tag pRb complex was found in all of 15 Tag positive cases.Conclusions:SV40 Tag was expressed highly in human brain tumors,and can form a specific complex with pRb,the inactivation of pRb due to the formation of Tag pRb complex may be an important mechanism in the etiopathogenesis of human brain tumors.
