摘要
目的:建立测定血浆中罗红霉素浓度的HPLCEC检测法,并与微生物法的测定结果相比较,评价两种方法的相关性。方法:以克拉霉素为内标,C18反相色谱柱,流动相为甲醇乙腈0.06mol·L-1磷酸二氢钠0.003mol·L-1EDTA(0.11∶0.39∶0.49∶0.01,pH7.0),安培型电化学检测器的工作电压+1.2V;0.5ml血浆样品经液液萃取纯化后溶于流动相中,进样20μl。结果:本法的线性范围0.1~10mg·L-1,日内RSD<3.2%,日间RSD<5.5%,方法回收率99.5%~103.3%,最低检测限0.1mg·L-1。比较HPLC和微生物法的测定结果,相关性显著(P<0.01)。结论:方法灵敏度高,重现性好,操作简便,适用于罗红霉素的药代动力学研究。
BJECTIVE: To establish a HPLC method for the determination of roxithromycin in human plasma using electrochemical detection,and to compare the results measured by HPLC with that by microbiological assay for assessing the correlation of the two methods. METHODS: Clarithromycin was used as the internal standard.The mobile phase consisted of CH3OHCH3CNNaH2PO4EDTA (0.110.390.490.01) and was adjusted to pH 7.0.The applied potential of the amperometric detection was set at +1.2v and the extracts of samples were chromatographed on a C18 reversedphase column after liquidliquid extraction. RESULTS: The method was linear over the range 0.110mgL-1.The intraday RSD was less than 3.2% and the interday RSD less than 5.5%.Method recovery was 99.5%103.3% and the limit of quantification was 0.1mgL-1.Analytical results for roxithromycin in plasma samples (n=280) between the two methods was found to be correlated lineal (P<0.01).
