碱法水解黑鱼鱼鳞及制备多肽螯合钙工艺的研究 被引量:16

Study on the hydrolyzation of snakehead scales with alkali method and the preparation of polypeptide-Ca^(2+)

摘要 采用碱法水解黑鱼鱼鳞制备多肽,并对其进行钙螯合修饰以制备多肽螯合钙。采用正交实验优选鱼鳞的最佳水解条件为:液比1∶10,氢氧化钠浓度1.0 mol/L,温度50℃,水解时间6h。电泳分析结果显示,水解鱼鳞多肽的分子量主要集中在6.5 ku。以螯合率为考察指标,通过单因素实验确定了多肽螯合钙的最佳制备条件为:多肽/CaCl2质量比4∶1,pH9.0,温度30℃,时间20 min。此条件下,螯合产物溶解度在pH1.0时最大,达3.95 g/100 mL。红外光谱结果表明,螯合产物在1411.57 cm-1处出现了Ca-NH2伸缩振动峰,而在1450.16 cm-1处的C=O面内摇摆吸收峰和3071.35 cm-1处的N-H收缩振动峰消失,表明多肽螯合钙的生成。 Polypeptides,which were extracted from snakehead scales by means of alkaline hydrolysis,were treated with chelating agent(Ca2+) to prepare polypeptide-Ca2+.The optimum hydrolysis conditions were indicated by orthogonal tests as follows: solid to liquid 1∶10(w/v),concentration of sodium hydroxide 1.0 mol/L,hydrolyzing temperature 50 ℃,hydrolyzing time 6 h.The results of electrophoresis analyzing showed that the molecular weight of most hydrolysates was about 6.5 ku.With chelate ratio as the index,the optimal preparation conditions for polypeptide-Ca2+ were obtained as the ratio of polypeptide to calcium 4∶1(w/w),pH 9.0,temperature 30 ℃,time 20 min.Under this condition,the best solubility(3.95 g/100 mL) for chelate products was obtained under pH 1.0.As seen from the infrared spectra,a new peak at 1115.93 cm-1 derived from stretching vibrations of Ca-NH2 of chelate products was observed;the peak derived from rocking vibration of C=O at 1450.16 cm-1 and stretching vibrations of N-H at 3071.35 cm-1 disappeared,indicating the formation of polypeptide-Ca2+.
