摘要
目的建立志贺菌肠毒素1基因(set1B)、肠毒素2基因(sen)和侵袭性质粒抗原H基因(ipaH)的三基因PCR检测方法,实现志贺菌三基因同时检测。方法选择志贺菌的3种毒力基因作为靶基因,在一个反应管中同时进行PCR扩增,根据产物分子量定性判断结果。结果三基因同时检测与单基因PCR分别检测3种毒力基因两种方法具有相同的灵敏度与特异性。62株志贺菌具有3种毒力基因型。结论三基因PCR检测志贺菌毒力基因具有简便、快速的特点,适用于医院实验室作为快速诊断志贺菌的辅助技术。
Objective A three-gene PCR method is established to detect Shigella enterotoxins 1 gene(set-1B),enterotoxins 2 gcne(sen) and the invasion ptasmid antigen H gene(ipaH) in one reaction.Methods three kinds of virulence gene of Shigella were selected as target genes and simultaneous amplified in one tube,and the qualitative judgments is based on the product molecular weight.Results The three-gene PCR and three single-gene PCR method to detect three kinds of virulence gene of Shigella have the same sensitivity and specificity.Conclusion It is convenient and fast to determine virulence genes of shigella with three-gene PCR,and may be used in clinic diagnose of Shigella.
