一株抗青枯假单胞菌放线菌的筛选、鉴定及发酵条件优化 被引量:8

摘要 从河南中牟土壤中筛选出一株对青枯假单胞菌(Pseudomonas solanacearum)有较强拮抗作用的放线菌ZM-16,它对几种常见病原细菌和真菌都有较强的抑制效果,其发酵液抗菌谱较广.通过菌株形态特征、培养特性、生理生化特性、化学特征研究以及16SrRNA序列分析,初步确定菌株ZM-16为链霉菌属的抗生链霉菌(Streptomyces antibioticus).通过发酵优化实验,确定该菌株的最佳发酵配方为:蔗糖8%、大豆粉5%、NaNO31%、NaCl0.2%、CaCO30.4%、K2HPO40.08%;最佳发酵条件为:发酵培养基初始pH值7.0,30℃,恒温摇床200r/min,培养6d,抑菌效果最好. An actinomycete strain ZM-16 with strong inhibitory effect on pathogenic eubacteria Pseudomonas solanacearum was isolated from soil in Zhongmou County,Henan,China.This study showed that the growth of some pathogenic bacteria and fungi was strongly inhibited by strain ZM-16 on plates.Its fermentation filtrate was found with broad antimicrobial spectrum.From the polyphasic taxonomical view,based on the study on its morphological,physiological and biochemical characteristics,together with sequencing of 16S rRNA genes and phylogenetic analysis,strain ZM-16 was initially identified as Streptomyces antibioticus.The fermentation condition of the strain was optimized.Its most effective fermentation conditions were obtained as cultivation for 6 days at 30 ℃ and 200 r/min,with initial pH of 7.0;and its optimum fermentation medium contained(w/V):8% sucrose,5% soybean meal,1% sodium nitrate,0.2% sodium chloride,0.4% calcium carbonate and 0.08% potassium phosphate.
