摘要
目的探讨加贝酯及核因子(NF-κB)在肝缺血再灌注损伤中的作用。方法采用Pringle’s法建立大鼠肝脏缺血再灌注模型,将40只SD大鼠随机分成四组,每组10只:假手术组(A组)、缺血再灌注组(B组)、加贝酯处理组(C组)、盐水对照组(D组)。光镜观察肝脏组织学变化,分别测定血浆中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)及组织中髓过氧化物酶(MPO)含量;并采用SABC免疫组化法观察肝组织中NF-κB的变化。结果 :A组肝组织形态正常,无NF-κB活化,肝功能酶学和MPO正常水平;B、D组肝细胞索排列紊乱,肝小叶变形,肝细胞和内皮细胞普遍水肿变性,中央区局灶性肝细胞坏死,汇管区中性粒细胞浸润,血窦内微血栓形成,NF-kB活化最为明显,血清酶学和MPO升高最为显著(P<0.01);C组能明显减轻这种损伤,NF-κB呈中度活化。结论肝缺血再灌注时,NF-κB被活化,使中性粒细胞向组织浸润,对肝脏缺血再灌注损伤起着重要的作用。
【Objective】To investigate the effect of gabexate mesilate(GM) and NF-κB on hepatic ischemia reperfusion injury in rats.【Methods】Models of rats with hepatic ischemia reperfusion injury were established with the Pringle's method.Forty SD rats were randomly divided into four groups: normal control group(group A),model group(group B),gabexate mesilate group(group C) and saline group(group D).Plasma alanine aminotransferase(ALT),aspartate aminotransferase(AST) and myeloperoxidase(MPO) in liver tissue were respectively measured in each group.The activity of NF-κB was determined by sABC immunohistochemistry,also histological examination of the liver was made.【Results】In group A,the histological test,ALT,AST and MPO were almost normal,NF-kB didn't have activity.In group B and D,hepatocytes were arranged in turbulence,hepatic lobules were distorted,hepatocytes and endothelial cells underwent swelling or necrosis,and neutrophils infiltrated in portal area,congestion and micro-thrombosis appeared in liver sinusoids;the activity of ALT,AST,MPO and NF-kB increased remarkably(P 0.01);but the injury was markedly decreased in rats pretreated with GM.【Conclusion】Nuclear factor kappa B is involved and may play an important role in liver ischemic reperfusion injury.
