摘要
目的 探讨顺铂诱导胃癌细胞凋亡过程中细胞内游离Ca2+ 浓度([Ca2+]i) 和三磷酸肌醇(IP3) 浓度的变化特点和意义。方法 选择顺铂诱导胃癌细胞凋亡,采用光镜、电镜、琼脂糖凝胶电泳和流式细胞仪检测凋亡。Fura2 荧光负荷技术测[Ca2+]i,竞争性蛋白结合法测定IP3 浓度。结果顺铂浓度为2 μg/ml 作用于BGC823 胃癌细胞24 小时出现典型的凋亡改变。胃癌细胞凋亡过程中,胞内[Ca2+]i 明显升高,以凋亡早期更为明显。胃癌细胞凋亡早期(30 秒),IP3 浓度明显升高,到凋亡晚期(2 ~24 小时) ,IP3 浓度明显降低。结论 在胃癌细胞凋亡过程中,[Ca2+]i 表现为上调趋势。IP3 浓度在凋亡早期,表现为上调趋势,在稍晚阶段,表现为下调趋势。
Objective To study the change and significance of intracellular free calcium levels ([Ca 2+ ]i) and inositol 1, 4, 5 trisphosphate (IP 3) levels in the process of gastric carcinoma cell apoptosis induced by cisplatin. Methods Apoptosis induced by cisplatin in gastric carcinoma BGC823 cell was investigated by light and electronmicroscopy, agarose gel electrophoresis and flow cytometry. [Ca 2+ ]i was determined by Fura 2 fluorescin load technic, IP 3 was determined by competitive protein binding method. Results When BGC823 cells were treated with cisplatin (2 μg/ml) for 24 hours changes appeared typical of apoptosis. [Ca 2+ ]i and IP 3 were significantly increased, especially at the initial stage of apoptosis. However, from 2 to 24 hours after cisplatin treatment, IP 3 levels progressively decreased, being significantly lower than those of the untreated controls. Conclusion In the process of cisplatin induced apoptosis of gastric carcinoma cells, [Ca 2+ ]i and IP 3 levels are up regulated at the very initial stage but down regulated from 2 hours afterwards.
