摘要
目的研究白细胞介素12(IL-12)与新城疫病毒(NDV)联合治疗小鼠S180肉瘤的效果及其作用机理。方法用NDV、IL-12、NDV+IL-12联合分组免疫治疗BALB/c小鼠S180肉瘤模型,通过分析瘤指数和抑瘤率及T细胞亚群分布、CTL活性,探讨免疫治疗效果与T细胞亚群分布及CTL活性之间的关系。结果NDV、IL-12、NDV+IL-12实验组的抑瘤率分别为(30.26±9.25)%、(31.85±7.24)%、(48.96±3.80)%,NDV+IL-12联合组的抑瘤率与NDV、IL-12单独治疗组相比,差异有显著性(P<0.01);瘤指数分别为(5.26±0.49)、(5.51±1.35)、(3.95±0.49),明显低于生理盐水组(7.67±0.67)(P<0.01),NDV组、IL-12单独治疗组与NDV+IL-12组小鼠相比,差异具有显著性(P<0.01)。NDV、IL-12和NDV+IL-12组的CTL活性明显高于生理盐水组(p<0.01);NDV+IL-12组又显著高于NDV、IL-12组(P<0.01)。NDV、IL-12、NDV+IL-12组的荷瘤小鼠外周血CD3+T、CD4+T、CD8+T细胞所占百分数均高于生理盐水组(P<0.01),且CD4/CD8比值也在升高。结论NDV、IL-12单独应用均能提高机体抗S180肉瘤的免疫力,NDV与IL-12联合免疫治疗能协同提高抗肿瘤作用,但其协同抗肿瘤机制还有待进一步研究。
【Objective】To investigate the anti-tumor effect of the combination of Newcastle disease virus (NDV) and interleukin 12 (IL-12) in treatment of murine subcutaneous tumor.【Methods】S180 cells (2×106) were injected subcutaneously into the murine left hind limb to establish the subcutaneous tumor model. The tumor-bearing mice were divided into four groups equally and randomly: NS control group, NDV treatment group, IL-12 treatment group and NDV + IL-12 treatment group. NS, NDV, IL-12 and NDV+IL-12 were respectively given to four groups after the tumor cells were inoculated for 7, 14 and 21 days. Ocular blood was obtained from mice in each group on the 28th day to analysis the CD3+T, CD4+T and CD8+T of peripheral blood, and the ratio of CD4+/CD8+ with FCM. Then these mice were executed with cervical dislocation to take spleen cells for testing the activity of CTL cells, to dissect tumor tissue completely for weighting and calculating the inhibition rate and index. 【Results】The tumor inhibition rates in NDV group, IL-12 group and NDV+ IL-12 group were (30.26±9.25) %, (31.85±7.24) % and (48.96±3.80) %. The tumor inhibition rates in NDV+IL-12 group were significantly higher than those in NDV and IL-12 group (P 0.05). Tumor index in NS control group was (7.67±0.67). Tumor indexes in NDV group, IL-12 group and NDV +IL-12 group were (5.26±0.49), (5.51±1.35) and (3.95±0.49), which were significantly lower than that in NS control group (P 0.05). Compared with NDV + IL-12 group, tumor indexes in NDV group and IL-12 group have significant difference (P 0.01). Compared with NS control group, the CTL activities in NDV group and IL-12 group have significant differences. The CTL activity in NDV + IL-12 group was higher than those in NDV group and Il-12 group (P 0.01). The CD3+T, CD4+T, CD8+T cells in mice blood in NDV group, IL-12 group and IL-12 + NDV group were all higher than those in NS control group. After separate NDV and IL-12 treatments and NDV + IL-12 treatment, these indexes were markedly improved, and the CD4+/CD8+ ratio also increased, the immune function of mice was strengthened. 【Conclusion】NDV and IL-12 can enhance the anti-tumor immunity of mice, IL-12 and NDV has synergistic effects in anti-tumor immune response and the detailed synergistic anti-tumor mechanism needs to be studied further
