微切变—助剂互作技术与超微粉碎技术加工的鹿茸粉对小鼠生长繁殖性能的影响 被引量:2

摘要 应用微切变—助剂互作技术和超微粉碎技术加工鹿茸获得鹿茸微切助粉和超微粉,采用有机溶剂索式提取法和热水浸提法提取鹿茸微切助粉和超微粉中的雌二醇、孕酮和睾酮性激素,用放射免疫法分析微切助粉和超微粉中性激素的溶出量;将鹿茸微切助粉和超微粉以2g/kg的比例分别添加到粉末状商品鼠粮中混匀,重新压制成棒状鼠粮,连续饲喂昆明小鼠9周。通过分析其对小鼠生长繁殖性能的影响,评价了微切变—助剂互作技术和超微粉碎技术加工鹿茸的应用效果。结果表明,运用微切变—助剂互作技术提取的鹿茸微切助粉中的雌二醇、孕酮和睾酮的溶出量均高于超微粉碎的鹿茸粉,尤其是水作为溶剂提取的雌二醇的溶出量是超微粉碎的15.5倍。饲喂含鹿茸微切助粉鼠粮的小鼠与饲喂含鹿茸超微粉鼠粮的小鼠相比,雄鼠的精子数、a级精子数和精子活力均高,具有统计学意义(p<0.05);卵巢和子宫的发育指数明显高于超微粉组,动情周期也比超微粉组短,血清中雌二醇、孕酮和睾酮性激素浓度显著增加(p<0.01);孕鼠的胚胎数量较多,发育较快,哺乳期仔鼠的生长也较快。以上结果充分证明,应用微切变—助剂互作技术加工鹿茸,能够高效释放目标活性物质。 The powder of deer antler velvet was prepared using auxiliary agents-mediated micro-shear technique(micro-shear powder) and ultra-fine comminution technique(ultra-fine powder).Estradiol,progesterone and testosterone were extracted by organic solvent and water from the two kinds of powder and quantified using radioimmunoassay techniques.Meanwhile,micro-shear powder and ultra-fine powder were mixed with the diet at a dose of 2 g/kg as an additive and fed to Kumming mice for 9 weeks.The sperm number and mobility,estrous cycle,litter size,gonad development and the final number of embryos were measured as indicators to analyse the difference in release efficiency of active substances between micro-shear powder and ultra-fine powder.Results showed that the dissolving-out quantity of estradiol,progesterone and testosterone from micro-shear powder was higher than that from ultra-fine powder.Water-extracted estradiol from microshear powder was 15.5 times as much as that extracted from ultra-fine powder.Significantly higher number of sperm and grade A sperm and higher sperm motility were observed in the group treated with micro-shear powder(P 0.05) compared with the mice treated with ultra-fine powder and the control group.Moreover,elevated sex organ index,shorter estrous cycle,higher level of sex hormone in serum,faster growth,and higher survival rate were also observed in the mice treated with micro-shear powder(P 0.01).This suggests that the auxiliary agents-mediated micro-shear technology is more efficient in release of active substances from deer antler velvet than traditionally used ultra-fine comminution technique.
