大鼠脑缺血再灌注区ICAM-1表达与白细胞浸润的观察及丹参的影响 被引量:40

摘要 目的 观察细胞间粘附分子( I C A M1) 蛋白在大鼠脑缺血再灌注的不同时程脑组织中的表达与中性白细胞浸润程度的关系及丹参对它们的影响。方法  S D大鼠分为3 组:假手术组、对照组及丹参组。大脑中动脉缺血2 h 再灌注2 h 、12 h、24 h 、48 h 、72 h 、7 d、14 d 后,分别进行 I C A M1 免疫组织化学及组织 H E染色。结果 在脑缺血再灌早期,脑微血管内皮细胞 I C A M1 免疫反应开始逐渐增加,再灌注48 h 达到高峰,再灌注14 d 接近正常水平,同时脑缺血区中性白细胞浸润也随之增加,在时程上与 I C A M1 表达同步。丹参组,再灌注48 h 后, I C A M1 免疫阳性血管数及中性白细胞的浸润比同时间对照组明显降低。结论 脑缺血 I C A M1 的表达与中性白细胞浸润密切相关,丹参能降低 I C A M1 的表达,抑制中性白细胞的浸润。 s Objective To investigate the correlation between the expression of intercellular adhesion molecule 1(ICAM 1) and neutrophil infiltration and the influence of Radix Salviae Miltiorrhizae(RSM) in cerebral focal ischemic reperfusion rats.Methods Immunohistochemical and histological techniques were used to evaluate ICAM 1 expression and neutrophil infiltration in ischemic brain tissue at different reperfusion time after middle cerebral artery occlusion (MCAO) for 2h in rats, and RSM (15 g/kg) was given peritoneally after MCAO followed by 48h reperfusion.Results There was a significant increase in ICAM 1 expression in the ischemic tissue that peaked at 48h reperfusion. Neutrophil infiltrations in ischemic tissue also increased after reperfusion and paralleled the increase of ICAM 1 expression. The expression of ICAM 1 and neutrophil infiltration in RSM treated rats decreased more significantly than in control rats. Conclusions These data indicate that the expression of ICAM 1 was significantly correlated with the degree of neutrophil infiltration in ischemic brain tissue, and RSM might inhibite the ICAM 1 expresion and neutrophil infiltration.
