高表达蛋白酪氨酸磷酸酶-PEST促进HepG2肝癌细胞体外增殖

摘要 为深入探讨蛋白酪氨酸磷酸酶-PEST在肝癌中的作用,构建了真核重组表达质粒pcDNA3.1/PTP-PEST并转染HepG2肝癌细胞,经荧光定量RT-PCR和Western blot对转染细胞进行筛选,选出稳定高表达PTP-PEST基因的细胞株,并采用MTT及Western blot对高表达PTP-PEST基因的细胞株的增殖速度和生长因子受体结合蛋白2表达量进行检测,观察到高表达PTP-PEST的细胞株内Grb2蛋白表达上调,细胞增殖速度加快.本研究表明,在HepG2肝癌细胞株内,PTP-PEST可能通过影响Grb2蛋白的表达来促进细胞增殖. Protein tyrosine phosphatase-PEST over-expression increases growth of HepG2. To investigate the function of protein tyrosine phosphatase-PEST (PTP-PEST) on hepatocellular carcinoma, the PTP-PEST gene was subcloned into pcDNA3.1(-) expression vector to obtain recombinant pcDNA3.1/PTP-PEST. HepG2 cell line was transfected with the recombinant vectors. The expression levels of PTP-PEST mRNA and protein were analyzed by real time RT-PCR and Western blot, and then the over-expression of PTP-PEST cell lines was filtered. After detection of cell growth and Grb2 expression by MTr and Western blot, the growth rate and level of Grb2 ascended in the over-expressed PTP-PEST cells. All the results indicate that PTP-PEST promotes HepG2 cell growth by regulating the content of Grb2.
