摘要
目的研究皮肤接触丙烯酰胺对雄性小鼠的生殖毒性作用。方法 36只健康雄性小鼠,随机分为阴性对照组,10、20和40 mg/kg剂量组共4组,每组9只。连续染毒35 d,每周测体重变化情况。单细胞凝胶电泳(SCGE)检测睾丸细胞DNA损伤,睾丸早期精细胞微核实验(MN)检测微核出现率。结果 不同剂量丙烯酰胺染毒后小鼠体重、睾丸重量与对照组相比呈下降趋势,睾丸细胞DNA损伤各指标与对照组相比,差异有统计学意义。各染毒组小鼠早期精细胞微核率高于对照组,20和40 mg/kg剂量组差异有统计学意义(P<0.01)。结论在本试验条件下,经过35 d皮肤接触丙烯酰胺可以对雄性小鼠生殖细胞产生毒性作用,而且随着接触剂量的增加,丙烯酰胺的生殖毒性作用越大。
Objective To study,after skin exposure of acrylamide,the reproductive toxicity in male mice.Methods The thirty-six healthy male mice were randomly divided into four groups,including control,10 mg/kg,20 mg/kg and 40 mg/kg group.Successively exposure for 35 days,body weight was observed every week.Single cell gel electrophoresis experiment was used to detect testicular cell DNA damage,early testicular sperm cell micronucleus test was used to detect the rate of micronuclei.Results The body weight,testis weight showed a downward trend when compared with the control group after different doses of acrylamide exposure.Each index of testicular cell DNA damage compared with the control group,the difference was statistically significant.The mice exposed to acrylamide had a higher rate of micronuclei than the control group,20 mg/kg and 40 mg/kg group had significant difference when compared with the control group(P 0.01).Conclusion Skin exposure to acrylamide can produce toxic effect on germ cells in male mice after 35 days,and with the exposure dose increasing,the reproductive toxicity of acrylamide become greater.
