冷暴露过程中大鼠骨骼肌组织氧化应激酶系的变化

摘要 目的探讨冷暴露过程中大鼠骨骼肌组织氧化酶系的变化。方法将48只大鼠分为6组,每组8只:对照组、冷暴露1、3、7、14、21天组,分别于室温饲养7天和0±4℃饲养1、3、7、14、21天,取大鼠小腿骨骼肌组织检测丙二醛(MDA)和抗氧化酶:铜锌超氧化物歧化酶(CuZnSOD)、锰超氧化物歧化酶(MnSOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽S-转移酶(GST)。结果大鼠骨骼肌组织MDA含量在冷暴露1~14天明显高于对照组(P<0.05,P<0.01),在冷暴露21天恢复正常;而CuZnSOD、MnSOD、CAT、GSH-Px、GST的活性从冷暴露第3天开始明显升高(P<0.05),CuZnSOD活性在冷暴露14~21天恢复正常,其他抗氧化酶活性在整个冷暴露期间均明显高于对照组(P<0.05,P<0.01)。结论冷暴露过程中大鼠骨骼肌抗氧化防护作用明显增强。同时,在冷暴露过程中随着骨骼肌抗氧化防护反应的增强以及MDA含量的恢复,可能标志机体冷适应的建立。 Objective To study the changes of enzymes of the oxidative stress in skeletal muscles of rats during cold exposure. Methods Forty - eight healthy male Wistar rats were randomly divided into 6 groups with 8 rats in each: control group, 1 - day cold exposure group, 3 - day cold exposure group, 7 - day cold exposure group, 14 - day cold exposure group and 21 - day cold exposure group. The contents of malondialdchyde (MDA) and activity of some antioxidant enzymes were determined in the skeletal muscle on day 1,3,7,14 and 21 after cold exposure. Results The results showed that comparing with control group, the level of MDA was obviously increased on day 1,3,7 and 14 after cold exposure and recovered on day 21 after cold exposure. The Copper,Zinc superoxide dismutase (CuZnSOD) activity was increased significantly on day 3 and 7 after cold exposure but returned to the control level on day 14 and 2I after cold exposure. In contrast,the activities of Manganese Superoxide Dismutase (MnSOD), Catalase(CAT), Glutathione Peroxidase (GSH -Px) and Glutathione S - transferase(GST) were obviously increased during the whole period of cold exposure. Conclusion The role of antioxidative defence was obviously increased in the skeletal muscles of rats during cold exposure. These data strongly suggest the establishment of cold acclimation on day 21 after cold exposure in rats.
